10.After equilibration, the Torrent will pause for sample
injection. Draw 2 mL of the dissolved Test Mix B solution
into a syringe and inject it:
a. Place the syringe on top of the Liquid Injection
Accessory valve.
b. Open the valve by turning the handle so it is in line with
the tubing.
c. Inject the liquid.
d. Close the valve by turning the handle so that it is
perpendicular to the tubing. Remove the syringe.
A chase solvent forces the sample through the automated
injection valve.
11.Inject 10 mL of ethyl acetate as a chase solvent:
a. Draw 5 mL of ethyl acetate into the syringe and inject
this as described in the sub-steps of step 10 above.
b. Repeat step a for a second 5 mL injection (10 mL total).
12.Touch the OK button to continue the run.
13.While the Torrent is operating, observe the Fraction
Collector/Fractionation Valve operation. Peaks should be
cut and collected in tubes/containers. Waste fluids should
be diverted to the waste container.
14.When the Torrent finishes the run, review the resulting
chromatogram which should meet the following criteria
(refer to Figure 2-22):
• Baseline must not rise more than 0.1 AU during
separation.
• Height of Peak 2 must be higher than Peak 3.
• Retention time of Peak 2 (time that the top of the
second peak occurs) must be between 5.5 and 7.5
column volumes.
If the criteria is not met, repeat the verification steps. If the
results still do not meet the listed criteria, contact your
Teledyne Isco representative.
